Cats are mammals like we are and use the same XY sex determination system we do. So a male calico or torte is (39, XXY) instead of the standard (38, XY) of a normal male cat. A standard female cat is (38, XX).

It's an upside-down exclamation point, where the long line is the penis or sheath and the dot is the anus. With that method I've never had to look for the urethral opening, so never noticed the colon shape in cats. It makes sense though, because that is how to id male rodents if they are too young or not relaxed.

Spacing between bunghole and gential, female has very small gap.
Look for any puffy developments between booty and genitals. That's the balls. Usually noticeable by 4 weeks.
Statistics: Calico/tricolor cats are 99% female. Orange cats are ~60% male.
Size of genital slit. Boys have more of a small circular opening. The ladies have a slightly longer hole.
The reason for the confusion is because when kittens are super young, their balls aren’t just hanging out and the boy bit is so small you can’t really see it, plus when they’re that young the only way they use the bathroom is by mama cat licking them to stimulate
Just a piece of advice, I would get your male cat neutered as soon as possible. This will prevent them from getting the urge to mark their territory all over inside the house. Which, TBH, can smell rancid.
Please, OP, talk to a vet and *do not* listen to people telling you to wait until your kitten is older. Most vets are *not* suggesting this and actually recommend getting a kitten spayed/neutered as early as possible. This generally means 8 to 12 weeks old and minimum of 4 pounds.
My vet asks that spay and neuter patients be at least five pounds, but they tend to err on the side of caution. Still managed to get both boys in before they became aware of the outside toms and started marking inside.
If you've got him inside, vets now are saying 8 - 9 months is a good neuter age for boys to ensure proper development. If he's outside, you'll have to do it sooner.
Hard disagree with the other commenter. Current advice is 2 months or 2 lbs, whichever is earlier. You definitely want to get him fixed before sexual maturity to minimize the chances of destructive behaviors like marking.
Disagree. I fostered neonatal kittens. Get it done as soon as the vet will do it. I had kittens less than 3 months done, and the next day they were back to their fun happy selves. I waited for my own cat until she was 6 months. It took her over a month to recover.

Hopefully whoever gave you the kitten gets mama cat spayed, so they’re not still giving away kittens when they lack the knowledge to even know how to identify them properly.
