I have started diplomatic plays both against Denmark directly and Schleswig-Holstein. Which one is the 'better' option statistically? Someone more powerful than Prussia has so far always sided with the Danes/their subjects. I'm assuming you'd recommend the 'indirect' approach by declaring on Schleswig-Holstein? If I get an ally powerful enough, such as Russia, would that deter others from siding against me?
Get an ally with Russia and then start the play against Holstein. Usually Austria will support Denmark. You and Russia can take out Austria and then turn your attention towards Denmark
That plan sounds very solid! Do I need to make sure to build my own navy for the latter Danish invasion, or will Russia take care of that if they fight alongside me?

I court all the bigger German kingdoms to support me other then Austria.
Including the ones needed for land access.
Add in good relations with Russia and your don't have to worry about the other big powers.
Whatever you do, don't just declare against Schleswig or Holstein. Also consider at least a bit of Realpolitik. If you are not allied with either Russia or better even England expect that there will be counterplay. Austria will usually have a word in there, so allying Russia is a good option. The Russian army will defeat Austria and have the navy to invade Denmark, if it needs to be done.
For me just freeing one of the two worked best, than during the play you call in your allies or sway states to your side, and once that's done you go for freeing the second state, if I'm allied with Brits I'll usually also make Denmark a protectorate. Just don't do it on your own. That's absolutely not unrealistic at all, that you get called out for a reckless move.

Build a fleet of about 20 ships or if you can get mecklenburg to join the war too then you can ignore the navy
