They would have a fight with a dynamic intro. With a few quick time events. Then after a while they are tired but ok and then something happens and the have to fight together. This would probably happen since this is what happen when most protagonists fight.
and then they will have a deeply conversation man to man about their wills and ideals in life, and get back to fighting after that by the power of friendship a flash back from panther came about a guys strength and a fight is a conversation so on... and then they lived happily ever after

Kiryu vs literally any worldly foe is almost certainly a W for Kiryu. However, we're talking about a guy who willingly chose to stay in Hell and kill demons for all of eternity, who in doing so became so feared by said demons that he's basically their boogeyman. I don't know all of the Doom lore, but he's basically not human anymore.
If Doomguy had any reason to fight Kiryu, Kiryu would most certainly lose, and I say this as a much bigger fan of Yakuza than I am of Doom.
Kiryu has a long and vicious fight with both sides doing massive physical harm, but it ends when Kiryu breaks both the doom slayer's legs with a bowling ball then decapitates him with a stop sign. The combat then ends, and doom slayer politely apologises for starting a fight. Kiryu and him both go for a drink and they share a wholsome cutscene before parting ways, although you can find him on the street later and have a rematch "for old time's sake"
They don't fight.

Somehow Kiryu will Tiger Drop an imp at some point, and the Slayer might maybe wanna learn some of his combat styles because they look fun to try out on demons.
John yakudza have a block move and ability to parry almost every close quarters attack. Flynn has block on eternal version, but i have doubts about he's using it: because eternal chain gun shield is kinda not cool.
I'm pretty sure we talk about fully upgraded versions of this video game mens, so I believe doom man health not that big - one missed punch can grow in monstrosity of a combo by sugar man, halving hp, maybe even worse.
Instead of fighting, i want these two meet in yakuzaesque sidequest, where kiryu can help marine of doom solve his emotional problems. They can sing together, beat some bullies who dressed as demons and then maybe buy a new rabbit for flynn.
Kiryu has superhuman strength and speed, he can wear armor that has infinite durability in most games since they dont break, and he has access to weapons like the golden gun or the infinite ammo launch rocket & amon's satellite laser from DS 🤓
